چکیده

The discovery of synthetic antihistaminic substances has had a great influence on studies of the cellular defense mechanism. Experiments with antihistamines have permitted better understanding of the functioning of certain defence processes. These substances inhibit the formation of the first barrier against bacterial invaders, and also the phagocytic activity of fixed (8,9, 10) and mobile (4, 10, 11) cells of the reticuloendothelial system. They also inhibit the action of certain mediators which maintain important functions of the defence apparatus (12,13). Consequently, they aggravate acute and chronic bacterial infections (3,5,6). We have found (3) that synthetic antihistamines also have a deteriorating effect in experimental tuberculosis in guinea-pigs and mice. Later, we showed that a synthetic antihistamine (mepyramine maleate) had a restraining effect on the phagocytosis of tubercle bacilli by monocytes (10) ; and that, contrarily, histamine stimulated the monocytes to an increased phagocytic activity toward the same bacilli (4). We then turned our attention to murine leprosy, another disease in which the mononuclear -macrophages play an important role. The purpose of the present investigation was to ascertain whether a similar mechanism could be detected in the macrophage-parasite relationship in murine leprosy. The first observation was contrary to that which was found with tubercle bacilli: the antihistamine did not inhibit the phagocytosis of Mycobacterium leprae murium by monocytes of guinea-pigs. This negative result stimulated the experiments which are reported here, designed to determine the influence of histamine and antihistamine on the phagocytosis of BCG and M. leprae murium by the monocytes of guinea-pigs and Wiersing rats.

ENDOGENOUS RELEASE OF OPIATES BY REPETITIVE ELECTRICAL FIELD STIMULATION IN THE GUINEA-PIG AND RAT ILEAL LONGITUDINAL MUSCLE

The effect of repetitive electrical field stimulation and the response of the guinea-pig and rat ileal longitudinal muscle to single pulse stimulations was examined. Single pulse field stimulation produced twitch contraction which was inhibited by repetitive field stimulation (10 Hz, 40V, 0.5 msec for 5 m). This inhibition was largely, though never completely, reversed by naloxone. Contrac...

Relaxant effect of pioglitazone on the guinea-pig isolated trachea through the modulation of endogenous prostaglandins

Thiazolidinediones are commonly used anti-diabetic drugs. Owing to the anti-inflammatory action of TZDs as a result of there action on the PPAR gamma receptor and a proposed action on the prostaglandins, these drugs can be tried in the acute exacerbations of COPD that are also commonly found among diabetic patients. An experimental study of one week was carried out at animal house of Army Medic...

Suppression of severe airway inflammation with non-toxic dietary nutrients in histamine-challenged guinea pigs: A comparison with Ibuprofen

We hypothesize that selected combinations of nutrients commonly encountered in the human diet, suppress asthma-associated airway inflammation more effectively than currently available anti-inflammatory medications. Here we demonstrate that biflavones and terpenes, distributed widely in human diet (in this case derived from Ginkgo biloba) synergize with the carotenoid antioxidant astaxanthin and...

Antihistaminic Effect of Bunium persicum on Guinea Pig Tracheal Chains

In a previous study, the relaxant and anticholinergic (functional antagonism) effects of Bunium persicum (B. persicum ) have been demonstrated on guinea pig tracheal chains. To elucidate the other mechanisms responsible for this relaxant effect, the inhibitory effect of this plant on histamine H1 receptors was examined in this study. The antihistaminic effects of aqueous and macerated extracts,...
